EVALUATION OF A RAPID PHAGE AMPLIFICATION ASSAY FOR DETECTION OF RIFAMPICIN RESISTANCE IN MYCOBACTERIUM TUBERCULOSIS ISOLATES

The purpose of the study was to evaluate rapid phage amplification assay in the determination of rifampisin resistance in Mycobacterium tuberculosis. A total of 50 M. tuberculosis isolates from various samples of the patients with active tuberculosis (TB) out of which 25 isolates were resistant to at least rifampicin and 25 were susceptible to four major anti-TB drugs (isoniazid, rifampicin, ethambutol, streptomycin) were taken into the study. Susceptibilities of the isolates to anti-TB drugs were analyzed by resistance ratio and BACTEC 460 TB system. Rifampicin-resistant and rifampicinsusceptible isolates were analyzed by a commercially available rapid phage amplification test, FastPlaqueTB-RIF. Twenty-two isolates among 25 susceptible strains were found as susceptible by FastPlaque whereas three isolates were evaluated as intermediate. Twenty-four isolates out of 25 resistant strains were found as resistant by FastPlaque whereas one resistant isolate was evaluated as intermediate. When compared with conventional methods, sensitivity, specificity and overall accuracy of FastPlaqueTB-RIF were found as 96%, 88% and 92%, respectively. It is concluded that FastPlaqueTB-RIF test may be a useful tool for rapid and accurate detection of rifampicin resistance in M. tuberculosis.
