COMPARISON OF IN-HOUSE PCR WITH REAL-TIME PCR FOR THE DETECTION OF CYTOMEGALOVIRUS-DNA

Cytomegalovirus is a member of the Herpes family, causing severe disease in immunocompromised hosts. The aim of this study was to compere in-house PCR with real-time PCR for the detection of CMV-DNA in clinical specimens.

Twentyfive clinical specimens were included in the study. These specimens were 22 sera, one urine, one bronchoalveolar lavage and one sputum. Cytomegalovirus-DNA was detected in clinical specimens with in-house PCR (Maksim Biotech, Inc., USA) and real- time PCR (Fluorion, Iontek, Turkey). The results from two techniques were compared with McNemar chi square test for statistical analysis. Cytomegalovirus-DNA was found positive in four specimens both by in-house PCR and real-time PCR, in two specimens only by real-time PCR and in one specimen only by in-house PCR. Cytomegalovirus-DNA was found negative in 18 specimens both by in-house PCR and real-time PCR methods. The agreement between the methods were estimated as 22 (88%). There was no statistically significant difference between in-house PCR and real-time PCR (p>0.05). It was concluded that real-time PCR and in-house PCR methods are in aggrement for detection of CMV-DNA in clinical specimens.
